Chemotaxis of Human Leukocytes

چکیده
Leukotaxis may be defined as the directional migration of leukocytes toward a chemotactic stimulus and is believed to represent the means by which a massive influx of polymorphonuclear leukocytes occurs in an area of inflammation (30). The attraction of polymorphonuclear leukocytes is brought about by a series of effector substances involving complement components (27), bacterial culture filtrates (14, 28), extracts from the leukocytes themselves (6), and tissues infected with virus (25, 31). The possible role of drugs on the inflammatory system has received increasing attention. Martin et al. (18, 19) recently reported an inhibitory effect of tetracycline on leukocyte migration, which was discovered during a serial study of leukotaxis using human leukocytes and sera. The purpose of the present study was to extend earlier studies on the effects of antibiotics other than tetracyclines on the chemotaxis of human neutrophils.

Stimulation of neutrophil leukocyte chemotaxis by a cloned cytolytic enterotoxin of Aeromonas hydrophila.
A cytolytic enterotoxin produced by Aeromonas hydrophila, isolate SSU, has been cloned in our laboratory. This enterotoxin lysed rabbit red blood cells, destroyed Chinese hamster ovary cells, caused fluid secretion in rat ligated ileal loops, inhibited the phagocytic function of mouse phagocytes, and was lethal to mice when injected intravenously. In this study, the effect of this cytolytic ent...

Monocyte chemotaxis is severely depressed in patients with advanced tumors, but the cellular basis for this chemotactic defect is not known. Because the actomyosin cytoskeleton is thought to play a primary role in chemotaxis, we have employed flow cytometry to examine several aspects of the contractile machinery including myosin II, myosin light chain kinase (MLCK), actin, and cytoplasmic calci...
متن کاملEffect of Escherichia coli alpha-hemolysin on human peripheral leukocyte function in vitro.
To gain further evidence for the role of the Escherichia coli alpha-hemolysin in pathogenesis, its in vitro effects on human peripheral leukocyte function were studied. Leukocytes exposed to low doses of alpha-hemolysin responded with a marked chemiluminescence response, indicating activation of oxidative metabolism. This response was time and dose dependent. Pretreatment of leukocytes with dos...
متن کاملComparative effects of moxalactam and gentamicin on human polymorphonuclear leukocyte functions.
The in vitro effects of moxalactam and gentamicin on chemotactic, phagocytic, and microbicidal activities of human polymorphonuclear leukocytes have been studied. Incubation of cells with moxalactam (25 to 400 micrograms/ml) resulted in a 10 to 15% inhibition of leukocyte chemotaxis (P less than or equal to 0.10). Incubation of cells with gentamicin (5 to 40 micrograms/ml) inhibited chemotaxis ...
متن کاملA Neutrophil-immobilizing Factor Derived from Human Leukocytes
A factor has been derived from human leukocytes which irreversibly inhibits the response of human neutrophils to diverse chemotactic stimuli without impairing their viability. It is released by both polymorphonuclear and mononuclear leukocytes during incubation in acidic medium, after endotoxin exposure and subsequent incubation in low potassium medium, and during phagocytosis of particles. It ...
